ANTINEOPLASTIC AND ANTIOXIDATIVE EFFECTS OF A METHANOLIC EXTRACT OF GANODERMA LUCIDUM

Background : Ganoderma lucidum  (FR.) KARST. is a holy mushroom or lacquered mushroom and has been used in Oriental medicine for a long time as a health tonic and sedative, and for the ailment of hepatitis, chronic bronchitis, impotence, cancer etc. Recently, several pharmacological activities of ganoderma have been experimentally discovered including immunopotentiation, antiviral, hypoglycemic, platelet aggregation inhibition, antihepatotoxic and anticancer activities.
Methods : The dried fruiting bodies of G. lucidum  from the mushroom farm in Bangkok were extracted with methanol in ratio 1:10 (w/v). The methanolic extract was investigated for antineoplastic and antioxidative potentials. In the antineoplastic study, the extract was tested for cytotoxicity against a suspension cell line, murine lymphocytic leukemia (P388) and four monolayer carcinoma cell lines, human mouth epidermoid carcinoma (KB), human cervical carcinoma (HeLa), human breast carcinoma (MCF-7) and human colon adenocarcinoma (HT29) with viability dye exclusion staining and total cell protein determination. The results were expressed as survival curves and median effective doses (ED50). In the antioxidation study, the antioxidant capacity of the extract was measured by using the photochemiluminescence method (PCL). The result was presented in equivalent concentration unit of Trolox.
Results : The ED50 values of the methanolic extract of G. lucidum  towards P388, KB, HeLa, MCF-7 and HT29 cell lines were 38, 119, 161, 126 and 152 mg/ml, respectively. The antioxidant capacity of lipid soluble substances of methanolic extract of G. lucidum  equals to 330 mmol Trolox equivalent / g methanol extract.
Conclusion : The methanolic extract of G. lucidum  exhibited slightly cytotoxic on P388, KB, HeLa, MCF-7 and HT29 cell lines. However, the result from PCL method showed a high antioxidant capacity that may enhance the health benefits of the mushroom.
